Proglumide powder Introduction

Proglumide powder was the first discovered gastrin antagonist, and both it and benzotript are derivatives of glutaramic acid. Its molecular structure is similar to the terminal structures of two intestinal kinins, gastrin (G-17) and cholecystokinin (CCK). Therefore, its functional amide group specifically competes with G-17 for G-17 receptors on parietal cells, significantly inhibiting G-17-induced secretion of gastric acid and pepsin, increasing the content of aminohexoses in the gastric mucosa, promoting glycoprotein synthesis, and protecting the gastric mucosa, thereby improving the symptoms of peptic ulcers and promoting ulcer healing. This product has little inhibitory effect on gastric acid secretion induced by histamine and vagus nerve stimulation. It does not cause a rebound phenomenon in gastric acid secretion when treating peptic ulcers and gastritis, and gastric acid secretion can remain at normal levels for up to six months after discontinuation of treatment. Because its inhibitory effect on gastric acid secretion is weaker than that of H2 receptor antagonists, it is no longer used alone in the clinical treatment of peptic ulcers, but its choleretic effect has recently received more attention.

Application Areas

  • Peptic Ulcers: Proglumide was initially developed and is primarily used to treat gastric and duodenal ulcers. It works by inhibiting gastric acid secretion and reducing gastrointestinal motility.

  • Gastric Mucosal Protection: It helps protect the gastric mucosa and promotes ulcer healing.

  • Anti-tumor Potential: New research suggests that proglumide may possess anti-cancer properties. It can inhibit tumor growth and metastasis by increasing T-cell infiltration through altering the tumor microenvironment, such as reducing fibrosis.

  • Neuroscience: Proglumide is a selective CCK antagonist and is often used as an experimental tool to study the role of CCK as a neurotransmitter or neuromodulator in the central nervous system.

FAQ

Q1: What is the target receptor of Proglumide powder?

A1: Proglumide is a cholecystokinin receptor (CCKR) antagonist which non-selectively binds to both the cholecystokinin receptors (CCK1R and CCK2R).

Q2: How does Proglumide powder differ from other anti-ulcer products?

A2: Compared to H2 receptor antagonists, proglumide has a weaker inhibitory effect on gastric acid secretion, but it does not cause a rebound increase in gastric acid and has a low recurrence rate, making it particularly suitable for long-term treatment and prevention of recurrence.

Q3: What are the common side effects of Proglumide powder?

A3: Common side effects are usually mild and transient gastrointestinal discomfort, including nausea, vomiting, abdominal pain, constipation and diarrhea, headache, and dizziness.
