Exenatide is a peptide consisting of 39 amino acids and is a long-acting glucagon like peptide-1 receptor agonist with an IC50 value of 3.22 nM. It significantly increases NO production, endothelial NO synthase (eNOS) phosphorylation, and GTP cyclohydrolase 1 (GTPCH1) levels in human umbilical vein endothelial cells in a dose-dependent manner. It has cytotoxic effect on MCF-7 breast cancer cells, with IC50 of 5 μ M at 48 hours. Low dose and high-dose exendin-4 treatment in ob/ob mice improved serum ALT and reduced blood glucose, and HOMA scores were calculated compared with the control group. Animals treated with it showed more pancreatic acinar inflammation, more nuclear condensation, and significant weight loss compared to control rats. It is associated with lower leptin levels and lower HOMA values in rats.
