Intracellularly, GS-441524 is phosphorylated to nucleoside monophosphate (NMP) by cellular kinases, and then converted into an active triphosphate metabolite (NTP structural analog). In viral RNA synthesis, the active NTP structural analog acts as a competitor to natural nucleosides (ATP, TTP, CTP, GTP) in RNA transcription. When the GS-441524 molecule is inserted into the transcription product, transcription terminates prematurely, thereby inhibiting the viral RNA transcription process.
